内毒素介导的多脏器功能障碍综合征动物模型的研究 被引量:12

摘要 目的:建立简单有效的内毒素介导的多脏器功能障碍综合征(MODS)动物模型,观察此模型的血流动力学、血液生化和脏器功能等变化特点。方法:选取10头健康杂交家猪,经麻醉和气管插管后用标准内毒素(LPS)(0.25mg·kg-1)由右侧颈内静脉缓慢注入,分别于注入内毒素前0h及注入后1h、2h、3h、4h、5h、6h记录血流动力学指标,检测血常规、血生化及脏器功能指标。结果:静脉输注内毒素后,平均动脉压(MAP)较前显著下降(P<0.01),肺动脉压(PA)持续升高。凝血酶原时间(PT)延长,血清肌酐(SCr)和丙氨酸转氨酶(ALT)升高。内皮素-1(ET-1)先有轻度升高,随后开始下降,而可溶性血栓调节蛋白(sTM)的变化相反。结论:内毒素输注能够诱发猪脓毒血症,并在6h内成功建立猪MODS模型。 Objective:To establish an effective and simple animal model of endotoxin induced Multiple Organ Dysfunction Syndrome (MODS). Methods: Intravenous lipopolysaccharide (LPS, 0.25mg·kg^-1) was given through right internal jugular vein to ten white cross pigs which have been anesthetized and mechanically ventilated. Hemodynamics, blood routine, blood chemical measurement and function of organ were examined at basement (0h) and at 1h,2h,3h,4h,5h and 6h after injection respectively. Results: After LPS infusion, mean arterial pressure (MAP) significantly decreased at 6 h(P〈0.01), but pulmonary arterial pressure (PA) went on increasing.Prothrombin time (PT),Serum creatinine (SCr), and the alanine aminotransferase (ALT) remarkably increased at 6h. Endothelin-1 (ET-1) decreased after slightly increase, while the change of soluble thrombomodulin (sTM) was opposite. Conclusion: The animal model of MODS can be effectively duplicated by LPS in pigs in six hours.
