三草安前汤对慢性非细菌性前列腺炎模型大鼠IL-1β、IL-6及TNFRⅡ表达的影响 被引量:6

摘要 目的观察三草安前汤对慢性非细菌性前列腺炎模型大鼠炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)及肿瘤坏死因子受体Ⅱ(TNFRⅡ)基因和蛋白表达的影响,阐明中药复方治疗慢性非细菌性前列腺炎的炎症因子蛋白靶点。方法将72只健康雄性Wistar大鼠随机分为6组,每组12只,除正常组外,其余5组应用大鼠前列腺蛋白提纯液辅以免疫佐剂制备实验性慢性非细菌性前列腺炎大鼠模型,正常组、模型组予生理盐水,西药组予消炎痛,三草安前汤大、中、小剂量组分别予不同剂量的三草安前汤连续灌胃。30d后处死,采用免疫组织化学、荧光定量RT-PCR等方法检测炎症因子mRNA及蛋白表达。结果三草安前汤可显著降低慢性非细菌性前列腺炎模型大鼠炎症因子IL-1β、IL-6的mRNA及蛋白表达水平(P<0.01),且大、中剂量组作用优于小剂量组(P<0.05),大、中剂量组与正常组比较差异无统计学意义(P>0.05);三草安前汤各剂量组TNFRⅡ表达降低,未随剂量增加出现明显变化(P>0.05)。结论三草安前汤对慢性非细菌性前列腺炎模型大鼠异常的免疫功能有调节作用,降低炎症因子表达是其治疗慢性前列腺炎的重要机理。 Objective To observe the effects of Sancaoanqian decoction on expression of cytokine IL- 1β, IL-6 and TNF R Ⅱ in experimental autoimmune prostatitis rats, and explore the target protein of cytokine of Chinese herbal compound in treating chronic nonbacterial prostatitis. Methods Seventy-two male Wistar rats were divided randomly into six groups and 12 rats in each group. All groups were made experimental autoimmune prostatitis rat model by injecting SC purified prostate protein with FCA except normal group. The normal and model group were given saline, western medicine group was given indomethacin, large, medium and small doses decoction groups were treated with large, medium and small doses of Sancaoanqian decoction. All rats were sacrificed after 30 days. The mRNA and protein expression of cytokines were tested by methods of immunohistochemistry and fluorescent quantitation RT-PCR. Results Sancaoanqian decoction could reduce the mRNA and protein expression of IL-1β, IL-6 in experimental autoimmune prostatitis rats. There were no significant difference between large and medium doses decoction group, but had a better activity than small dose. The expression of TNF R Ⅱ were reduced in all doses groups and no difference among three doses of Sancaoanqian decoction. Conclusion Sancaoanqian decoction can regulate the immune function in chronic prostatitis rat model. Down-regulation of cytokines maybe one of important mechanisms in treating chronic prostatitis.
