摘要
目的:观察不同糖浓度的Krebs-Henseleit(K-H)液对大鼠离体心脏缺血/再灌注损伤的影响。方法:采用Langendorff大鼠离体心脏灌流技术制备心脏缺血/再灌注损伤模型,用不同糖浓度的K-H液灌流心脏,灌流结束后测定心肌组织中肌酸激酶(CK)、乳酸脱氢酶(LDH)、谷胱甘肽-过氧化物酶(GSH-PX)的活性以及一氧化氮(NO)、丙二醛(MDA)的含量。结果:1.5倍糖浓度组(3g.L-1)能显著提高大鼠心肌组织中LDH、CK、GSH-PX的活性(P<0.05),提高NO(P<0.05)、降低MDA(P<0.05)的含量。结论:1.5倍糖浓度组(3g.L-1)对大鼠缺血/再灌注心肌有保护作用,可能是通过增加心肌细胞膜稳定性、增强大鼠抗氧化酶系统功能、松弛血管平滑肌增加血供、抑制心肌缺血/再灌注损伤时的脂质过氧化损伤来实现的。
Objective: To observe the influence of different concentrations of Glucose in Krebs-Henseleit (K-H) liquid on isehemia/repeffusion injury in rats with isolated working hearts. Methods: The heart models of ischemia/reperfusion injury were built with Langendorff isolated perfusion technology. The different concentrations of Glucose in K-H liquid were used for reperfusion. At the end of the reperfusion, the activities of Creatine Kinase(CK), Lactate Dchydrogenase( LDH), Glutathione peroxidase(GSH-Px) and the concentrations of Nitric Oxide( NO), Malondialdehyde(MDA) in the myocardial tissue were examined. Results: The group of 1.5 times Glucose concentration showed an increase in the enzymatic activities of LDH, CK, GSH-Px and an increase in the concentration of NO and a decrease in the concentration of MDA in the myocardial tissue. Conclusion: The group of 1.5-times-Glucose-concentration has a protective effect on myocardial ischemia/reperfusion injury because it improves the stability of myocardial cell membrane, enhances anti-oxidative enzyme activity, improves blood supply by relieving blood vessel smooth muscle, restrains the lipid hydroperoxide injury when myocardial tissue suffers from ischemia/reperfusion injury.
