链脲佐菌素加膳食高脂高糖饲料诱导的2型糖尿病大鼠模型的验证 被引量:10

摘要 目的:验证链脲佐菌素(STZ)加高脂高糖饲料诱导的2型糖尿病大鼠模型的可靠性。方法:50只Wistar大鼠适应性饲养1w,随机分为两组,普通饲料组20只和高脂高糖组30只。普通饲料组以普通饲料喂养,高脂高糖组先喂以高脂高糖饲料6w,由尾静脉采血测空腹血糖(FBG)和胰岛素(FINS),并计算胰岛素敏感指数;然后按30mg/Kg腹腔注射1%的STZ溶液,1次/w,连续腹腔注射2次,1w后测血糖。以连续2次空腹血糖≥7.8mmol/L为造模成功,共25只。两组大鼠于高脂高糖组大鼠给予STZ后第2周、第4周测定空腹血糖和口服糖耐量。结果:与普通饲料组比较,饲养6w后高脂高糖组大鼠胰岛素水平升高(P<0.05),高脂高糖组大鼠胰岛素敏感指数下降(P<0.01);给予STZ后2w高脂高糖组大鼠空腹血糖、2h血糖较普通饲料组升高(P<0.01);给予STZ4w后空腹血糖、1h血糖、2h血糖较普通饲料组大鼠升高(P<0.01)。结论:STZ加高脂高糖膳食诱导的2型糖尿病大鼠血糖升高,口服糖耐量异常,出现了明显的胰岛素抵抗,符合2型糖尿病的诊断标准。这种模型可靠,理想,简单,费用低,容易广泛推广。 Objective:To validate the reliability of type 2 diabetes rat model induced by streptozotocin (STZ)plus high calorie and high sugar diet. Methods:50 Wistar rats were divided randomly into two groups:high calorie and high sugar diet group(H group,n=30)and common diet group(C group,n=20). The levels of fasting blood glucose and insulin were detected and insulin sensitivity index was calculated on the sixth week after feed. At the same time,rats in H group were injected intraperitoneally 0.1% STZ at 30 mg/Kg once a week for successive 2 weeks,then fasting blood glucose and glucose tolerance were detected on the second and fourth week after injection. Results:Compared with C group,serum insulin levels of rats were significantly increased(P〈0.05),insulin sensitivity indices of rats were significantly decreased (P〈0.01) in H group on the sixth week after feed; the levels of fasting blood glucose and 2 h postprandial blood glucose of rats were increased in H group on the second week after injection(P〈0.01)and the levels of fasting blood glucose, 1 h and 2 h post- prandial blood glucose of rats were increased in H group on the fourth week after injection (P〈0.01). Conclusion :This method of establishing animal model is simple, reliable with good feasibility.
