摘要
切取嘉兰(GloriosarothschildianaO’Brien)0.5mm左右的茎尖组织接种在Murashige&Skoog培养基上,在25℃、16h光照(1000lx)条件下培养1个月可以获得再生植株。继代培养后,这些再生植株可以在试管内形成块茎。再生植株的茎基部分生能力较强,利用其茎盘反复继代培养可以获得大量不定芽,再经过分株培养可以形成块茎,从而实现大量繁殖的目的。当培养基内的6-BA浓度提高到4mL/L时,可获得最多的不定芽和较大的茎盘。提高蔗糖浓度可以获得较大的块茎,当蔗糖浓度提高到9%时,块茎平均单重达0.48g。
In stem tip culture of Gloriosa rothschildiana in Murashige & Skoog medium at 25℃ and 16 hours light period (1000 lx) , we obtained adventitious shoots and regeneration plants, which formed tuber in vitro after subculture. The basal plate of regeneration plants exhibited a strong differentiation activity. A large number of regeneration plants and tubers were obtained through repetition of this stem base culture. The largest amount of adventitious shoots and big size of basal plate were obtained when 6-BA concentration was increased to 4mg/ L in culture medium. Moreover, bigger tubers were obtained when sucrose concentration in culture medium increased. Mean weight of tubers increased to 0.48g when sucrose concentration was increased to 9%.
