摘要
为研究可溶性白细胞表面分化抗原40配体(sCD40L)、细胞间黏附分子-1(ICAM-1)及甘油三酯(TG)在糖尿病下肢溃疡大鼠发病中的作用。采用雄性SD大鼠腹腔注射中等剂量链脲佐菌素(STZ)同时给予高脂饮食建立糖尿病模型;八周末,所有大鼠下肢涂抹冰醋酸建立糖尿病下肢溃疡模型,观察溃疡变化;12周末处死大鼠,测体重并取血,测空腹血糖、TG,酶联免疫法测sCD40L及ICAM-1。注射STZ后糖尿病大鼠出现明显多饮、多尿、体重减轻症状;糖尿病并溃疡组血TG、sCD40L及ICAM-1明显高于其他组;而正常对照组与溃疡对照组各项指标无明显差异;CD40L与ICAM-1之间呈显著正相关,而TG与两者之间无相关性。由此可知,sCD40L、ICAM-1、TG可能在糖尿病足溃疡的发生中起了重要作用。
Study the role of soluble CD40 ligand ( sCD40L), intercellular adhesion molecule-1 ( ICAM-1 ) and tri- glyeeride(TG) in the diabetic rat with lower extremity ulcer. SD male rats were injected with medium-dose strepto- zotocin(STZ) and at the same time given high-fat diet. At the 8th weekends, all rats'lower extremity apply acetic acid to estabish model of diabetic leguleers, then observe the ulcers. All rats were killed at the 12th weekend, meas- ure the weight and the blood for fasting glucose, TG; sCD40L and ICAM-1 by euzymelinked immunosorbent assay (ELISA). Diabetic rats have significant syndrome of polydipsia,polyuria and weight loss;TG,sCD40L and ICAM-1 were significantly higher in the diabetes with ulcers group than other groups, while the control group and the control group with ulcers have no difference, sCD40L and ICAM-1 was obviously correlative,TG has no obviously correla- tion with sCD40L andICAM-1, sCD40L,ICAM-1 ,TG may play important role in diabetic foot ulcers.
