5-氨基-2,4,6-三碘-1,3-苯二甲酰氯经N-酰化、酰胺化得到N,N'-双[2-羟基-(1-羟甲基)乙基]-5-[(2S)-(2-乙酰氧基)丙酰胺基]-2,4,6-三碘-1,3-苯二甲酰胺,再经酸性脱乙酰基制得非离子型X射线造影剂碘帕醇,总收率约78%。
Iopamidol was synthesized from 5-amino-2,4,6-triiodo-1,3-phthaloyl dichloride via N-acylation and amidation to give N,N'-bis E2-hydroxy- (1-hydroxymethyl) ethyl] -5- [ (2S) - (2-acetoxy) propionamido] -2,4,6-triiodo-1,3- phthalamide, which was subjected to acidic deacetylation with an overall yield of about 78 %.
