不育患者精子中肺癌肿瘤抑制因子-1的表达及意义探讨

Study on the significance of the expression of TSLC1 in the sperm of infertile men

摘要 目的探讨肺癌肿瘤抑制因子1(TSLC1)与男性不育的关系。方法首先免疫小鼠制备TSLC1单克隆抗体(McAb),然后分别选取30份正常组和40份不育组精液标本,Percoll梯度离心分成精浆和精子,将分离出的精子与制备出的McAb做免疫组织化学染色,对结果进行统计学分析。结果所有正常生育组精子的TSLC1蛋白表达均为阳性。在不育组中,30例弱精子症患者,25例TSLC1蛋白表达为阴性,10例少精子症患者,8例TSLC1蛋白表达为阴性。正常组与弱精子症组、少精子症组差异分别有统计学意义(P<0.001),弱精子症组与少精子症组差异无统计学意义(P>0.05)。结论精子中TSLC 1蛋白的低表达或不表达与男性不育密切相关。 Objective To explore the relationship between TSLC1 and male infertility. Methods TSLC1 monoclonal antibody was produced in immunized mice. Thirty normal semen specimens and fourty infertility semen specimen were collected. Seminal plasma and sperm were separated by Percoll gradient centrifugation. The expression of TSLC1 was detected in sperm by immunohistochemistry analysis. Results The expression of TSLC1 was detected in all samples of the normal group. In the infertility group, the expression of TSLC1 was negative in 25 of 30 asthenospermic patients and in 8 of ten oligospermia patients, no significant difference was found in the expression of TSLC 1 between asthenospermic patients and oligospermia patients(P=0.810). While there were significant differences in expression of TSLC1 between normal group and other groups(P=0.000). Conclusion The low level or null expression of TSLCI protein in sperm, might be associated with male infertility.
