摘要
目的通过对雄性新西兰兔精液中精子质量及受精能力的检测,观察睾丸大体形态及精子细胞超微结构的改变,探讨局部振动对雄性兔的生殖功能的影响。方法将新西兰兔随机分为A组(接振强度为3.02m/s^2)、B组(接振强度为6.13m/s^2)、C组(接振强度为12.26m/s^2)和1个对照组。于接振后30d测定精浆中精子的密度、活动力、活动率、畸形率,通过去透明带仓鼠卵-精子穿透试验和精子低渗膨胀试验评价精子的受精能力,光镜下观察睾丸组织学变化,电镜下观察睾丸精子细胞超微结构的改变。结果接振试验后与对照组相比较,A、B、C 3个试验组新西兰兔精液精子活动率、活动力明显降低(P<0.01);精子的畸形率明显升高(P<0.05)。精子的受精率及尾部肿胀的精子比率明显降低(P<0.05)。睾丸生精细胞数量减少,精子细胞的部分细胞器破坏、裂解、坏死。结论后肢接振试验可降低精子的活动力、活动率及卵子受精率及低渗肿胀率,增加精子的畸形率,睾丸组织中生精细胞减少,精子细胞的部分细胞器破坏、裂解、坏死,从而抑制精子的成熟,影响生殖功能。
Objective To study the effects of behind legs vibrations on reproduction of male rabbits through evaluation of sperm quality in seminal plasma,the histological morphology of testis,the zona-free hamster oocyte test (ZHOT) and hypoosmotic swelling test (HOS) and ultrastructure of sperm. Methods All male adult rabbits were randomly divided into four groups including group A (vibration power: 3.02m/s^2), group B (vibration power: 6.13m/s^2), group C (vibration power: 12.26m/s^2) and control group. The density, motility, viability and malformed rate of sperm in semen were measured and analyzed respectively, and then the oocyte penetrating rate and hypoosmotic swelling rate were assessed by zona-free hamster oocyte test (ZHOT) and hypoosmotic swelling test (HOS)after vibrated for 30 days. The histological morphology of testis and ultra structure of sperm were observed by light microscope and electron microscope. Results Compared with those of control groups, the motility, viability, the oocyte penetrating and hypoosmotic swelling rate of sperm were much lower in three test groups (P〈0.01), whereas the malformed rate were much higher (P〈0.05). The number of spermatogenic cells were decreased and some organelles of sperm were destructed. Conclusion All data in the study suggested that the vibration of behind legs of rabbits affected the motility, viability, the oocyte penetrating rate, hypoosmotic swelling rate in semen, the malformed rate of sperm, the number of spermatogenic cells and the structure organelle of sperm, which might inhibit the mature of sperm and further reduce the reproduction of male rabbits.
