摘要
研究表明,猪蛔虫三期幼虫、分泌排泄物等天然抗原对宿主能提供有效的免疫保护,一些猪蛔虫融合大肠杆菌重组表达蛋白也可作为防控猪蛔虫病的候选疫苗。作者已克隆了猪蛔虫rAs37基因并进行了原核表达,并通过对重组融合蛋白As37进行免疫原性分析,显示其具有良好的免疫学活性。在此基础上进行了动物免疫保护性试验,并利用大肠杆菌高效表达的重组蛋白,建立ELISA方法对血清抗体水平进行了检测,为开发蛔虫疫苗,进一步有效防治猪蛔虫及研究猪蛔虫的免疫学诊断方法打下了基础。
Prior studies have showed that pigs can be rendered immune to Ascaris suum infection by immunisation with crude antigen such as third stage larvae, and ES. Recently, some Escherichia coli-expressed recombinant antigen from A. suum were reported to be suitable vaccine candidates for the control of Ascaris infection. Here we cloned As37 gene by re- verse transeription-polymerase chain reaction (RT-PCR) using primers based on the pubmed eDNA sequence of As37 gene. The Escherichia coil-expressed recombinant protein showed highly immunoreactive by Western-blot analysis and immune protective assay. ELISA was established using recombinant protein As37. The result showed that it can be considered as a potential vaccine and diagnostic candidate.
