摘要
目的:探讨中等强度运动训练对心理应激大鼠行为和淋巴细胞热休克蛋白70表达的影响。方法:32只健康雄性SD大鼠随机分为4组:对照组、心理应激组、运动组和运动+心理应激组,每组8只。对照组不进行运动和心理应激干预,正常饲养;心理应激组前7周同对照组,第8周采用每天一次、每次2小时、持续3周的噪音刺激诱发建立心理应激模型;运动组每天无负重游泳训练1次,每次60分钟,共10周;运动+心理应激组运动10周,从第8周开始附加与心理应激组同样的应激。实验后测定各组大鼠旷场行为和外周血淋巴细胞热休克蛋白70表达。结果:(1)与对照组相比,心理应激组和运动组大鼠爬格块数均显著增加(P=0.024,P=0.047),心理应激组和运动组大鼠直立次数显著降低(P=0.03,P=0.001);双因素方差分析表明,运动训练和心理应激交互作用对大鼠爬格块数(F=6.30,P=0.02)和直立次数(F=6.91,P=0.01)有显著影响。(2)与对照组相比,心理应激组和运动+心理应激组大鼠热休克蛋白70表达均显著增加(P<0.01),运动组热休克蛋白70显著下降(P=0.04);双因素方差分析表明中等负荷运动训练和心理应激交互作用对大鼠淋巴细胞热休克蛋白70表达有显著影响(F=4.26,P=0.048)。结论:中等强度运动训练能减小心理应激导致的大鼠行为和淋巴细胞热休克蛋白70表达的变化,提示适宜强度的运动能够拮抗心理应激对机体造成的损伤。
Objective To study the effect of moderate exercise on the behaviors and heat shock protein 70 (HSP70) expression in peripheral blood lymphocytes of rats with mental stress. Methods Thirty two healthy male SD rats were randomly and equally divided into four groups: control group (C), 3-week noise-induced mental stress group(P), 10-week swimming group (S), 10-week swimming plus 3-week mental stress group (PS). Behaviors and HSP70 expression in the peripheral blood lymphocytes of rats with mental stress were determined. Two-way ANOVA and LSD analysis were applied for the statistical analysis. Results ( 1 ) As compared with the group C, the numbers of crossing in groups P and S significantly increased (P=0.024 and P=0.047, respectively); the numbers of rearing in groups P and S significantly decreased (P =0.03 and P =0.001, respectively), furthermore, there was significant interaction of moderate exercise and mental stress on the numbers of crossing and rearing (F=6.30, P=0.02 and F=6.91, P= 0.01, respectively). (2) As compared with the group C, the HSP70 expression in lymphocytes in groups S and PS significantly increased (P〈0.01 and P〈0.01, respectively), and in group P increased less (t=-3.59, P= 0.04). Significant interaction of moderate exercise and mental stress on the HSP70 expression in lymphocytes were also found (F=4.26, P= 0.048). Conclusion Moderate exercise could reduce the intensity of stress reaction.
