摘要
目的:探讨脂肪组织UCP2mRNA和TNF-α在运动防治高脂饮食危害中的作用。方法:SD雄性大鼠30只随机分为普通饮食对照组(C)、高脂饮食对照组(H)、高脂饮食运动组(HE)和普通饮食运动组(E)。H组和HE组大鼠采用高脂饲料喂养;HE组和E组同时进行12周无负重游泳运动(90min/天,5次/周)。采用RT-PCR方法测试大鼠脂肪组织中UCP2mRNA和TNF-αmRNA表达水平,采用ELISA法测定大鼠脂肪组织和血清中TNF-α的含量。结果:(1)12周实验结束后,H组体重显著高于HE组和C组,但脂体比与C组比较差异不显著。HE组脂体比显著低于H组和C组;(2)H组大鼠血清TNF-α含量显著高于HE组和C组;HE组脂肪组织TNF-α含量与H组和C组比较差异不显著,E组脂肪组织中TNF-α含量显著低于HE组和C组;(3)H组大鼠脂肪组织中UCP2mRNA和TNF-αmRNA表达显著高于HE组和C组;HE组脂肪组织中UCP2mRNA和TNF-αmRNA显著高于C组和E组;E组和C组之间差异不显著。结论:高脂饮食大鼠脂肪组织TNF-αmRNA和UCP2mRNA升高,运动可降低高脂饮食大鼠脂肪组织TNF-αmRNA和UCP2mRNA升高的程度,这种调节变化可能在运动减少高脂饮食导致的脂肪累积中发挥一定的作用。
Objective The purpose of this experiment was to investigate the effect of exercise on the UCP2 (uncoupling protein-2) and TNF-α (tumor necrosis factor alpha) in adipose tissue. Methods Thirty male SD rats were randomly assigned to four groups: control group(C); high fat diet group(H); exercise group(E); high fat diet in combination with exercise group(HE). The rats in groups H and HE were fed with high fat diet,and the rats in groups C and E were fed with standard rodent chow ad libitum. Rats in the group E underwent swimming for 12 weeks (90min/day, 5days/week). The UCP2 mRNA and TNF-α mRNA in adipose tissue were determined by RT-PCR. TNF-α concentration in adipose tissue and serum was tested by ELISA. Results (1) After 12-week feeding with high fat diet and high fat diet plus swimming, body weight of group H were higher than that of group E and group C, but there was no significant difference in ratio of fat to body weight between group H and group C. The ratio of fat to body weight of group HE was lower than that of groups C and H. (2)The serum TNF-α of group H was higher than that of groups HE and C. There was no significant difference in TNF-α concentration in adipose tissue between groups HE and H, but TNF-α of group E was significantly lower than that of group HE. (3)The level of UCP2 mRNA and TNF-α mRNA expressions in adipose tissue of group H was the highest, and of group HE was significantly higher than that of group E and group C, and no significant difference was found between group E and group C. Conclusion TNF-α mRNA and UCP2 mRNA in adipose tissue of rats fed with high fat diet increased,exercise lowered the increasing of TNF-α mRNA and UCP2 mRNA in adipose tissue of rats fed with high fat diet indicating that exercise played an active role in preventing excessive accumulation of fat induced by high fat diet.
