海门山羊发情周期血浆和发情后子宫内膜17β-雌二醇、孕酮及其受体水平的变化 被引量:1

CONCENTRATIONS OF 17β-ESTRADIOL, PROGESTERONE AND THEIR RECEPTORS IN THE PERIPHERAL PLASMA DURING THE OESTROUS CYCLES AND ENDOMETRIUM AFTER THE ESTRUS IN HAIMEN GOATS

摘要 13只海门山羊、年龄1-2岁,其中5只在发情周期的0(发情当天)-9、11、13、15、17、19、20天,每天08:00-09:00颈静脉导管采血1次。所得血浆置-20℃冰箱保存。另8只羊在采集血样的同时,于发情后2、3、4、5、6、7天,取出子宫置液氮中保存。分别用放射免疫法、H^317β-雌二醇和H^3孕酮交换法测定血浆、子宫内膜胞浆、胞核内17β-雌二醇(E_2)和孕酮(P)及其受体(E_2R、PR)水平。结果表明,发情前1天外周血浆E_2水平最高,为27.0941±3.9328pg/ml。发情当天为18.8265±1.3710pg/ml。发情后期波动在17-19pg/ml之间。黄体中期(5-13天)较低,平均16pg/ml左右。而血浆P水平则发情当天最低,为0.5289±0.0802ng/ml。发情后的整个黄体期不断升高,峰值出现在发情后的第17天,为6.0406±1.3516ng/ml。17天后急骤下降(p<0.001)。发情后2-3天。胞浆E_2和E_2R分别由2.8441上升到3.3646pg/mg蛋白和83.7005上升到259.9745pg/mg蛋白;P和PR水平,分别由0.0892ng/mg蛋白上升到0.2416ng/mg蛋白和168.8552上升到220.6367±39.3077ng/mg蛋白。3天后,虽血浆E_2水平上升,但胞质、胞核E_2及其受体水平下降,胞质PR水平亦下降,而胞核PR水平不断上升。胞质P水平起初下降,5天后与核PR水平同步上升(r=0.9444,p<0.05)。 Thirteen Haimen goats, 1 to 2 years old, were used in expriment. Blood samples of five goats were collected from the jugular vein at 08:00 or 09:00 AM every day from day 0(oestrus=day 0) to 9 and every other days from day 11 to 21 of the estrus cycle. The blood plasma was stored at -20℃. Uterus samples of another 8 goats were removed at 2 to 7 days alter oestrus and stored in liquid nitrogen. The plasma concentrations of 17β-oestradiol (E_2) and progesterone (P) and the endomctrical, cytoplasmic and nuclear levels of E_2, P and their receptors in were determined by RIA and H^3-E_2, H^3-P exchange method respectively. The results indicated that the plasma concentrations of 17βE_2 were 27.0941±3.9328pg/ml at day-1, 18.8265±1.3710pg/ml at day 0, 17-19pg/ml in metestrus and 16pg/ml during luteal phase (5-13). The level of P in the plasma was 0.5289±0.0802ng/ml at day 0. It was elevated progressively through the lutcal phase. The peak value of P 6.0406±1.3516ng/ml appeared at day 17 of the cycle, and then declined abruptly (p<0.001). 2-3 days after estrus, the concentrations of E_2 and E_2 receptor in the cytoplasma and nuclear were elevated from 2.8441 to 3.3646 and 83.7005 to 259.9745pg/mg protein respectively. The concentrations of P and P receptor were increased from 0.0892 to 0.2416 and 168.8552 to 220.6367±39.3077ng/mg protein respectively. Three days later, although the plasma concentrations of E_2 elevated, the levels of E_2 and their receptors in cytoplasm and nucleus decreased. The levels of P receptor in cytoplasm also declined, but the levels of P receptor in nucleus increased progrssively, The levels of P declined at first five days after estrus, and then increased in coincide with P receptor in nuclear (r=0.9444, p<0.05).
