摘要
目的:观察咳喘安胶囊的镇咳和祛痰作用及对哮喘小鼠肺NF-κB表达的影响,阐述其作用的可能机制。方法:采用小鼠气管酚红排泌实验观察咳喘安胶囊祛痰作用。以氨水诱导的咳嗽实验观察咳喘安胶囊的镇咳作用。采用卵清白蛋白激发小鼠哮喘,然后采用咳喘安胶囊进行干预,免疫组织化学染色方法观察NF-κB在小鼠肺组织中的表达变化。结果:咳喘安胶囊中、高剂量组能显著促进小鼠气管酚红排泌,而低剂量组则没有明显祛痰作用。咳喘安胶囊中、高剂量组对氨水诱导的咳嗽有明显的延长诱导时间和镇咳作用。各哮喘模型对照组小鼠肺支气管壁NF-κB面积及积分光密度值均明显高于正常组(P<0.01),各用药组小鼠支气管壁NF-κB面积及积分光密度值均显著小于模型对照组(P<0.01),且中药高剂量治疗组支气管上皮细胞及肺泡壁炎性细胞胞核阳性的细胞比例显著低于哮喘组(P<0.05)。结论:咳喘安胶囊有很好的镇咳祛痰作用。咳喘安胶囊能显著降低哮喘小鼠气道壁细胞内NF-κB的表达。提示抑制NF-κB的表达是咳喘安胶囊治疗哮喘的可能机制。
Objective: To evaluate the antitussive and expectorant effects of Kechuanan capsule, and its effect on NF-κB of asthma mice lung. To explain the possible mechanism. Method: Mice cough was induced by ammonia. The number of cough and cough incubation period were recorded to indicate the effect of Kechuanan capsule. The expectorant activities were tested by the amount of phenol red in pulmonic excretion secreted in mice. The asthma mice model was indused by OVA, and given Kechuanan capsule gastric perfusion, the NF-κB change of asthma mice lung was evaluated by immunohistochemistry. Results: Compared with the normal saline group, Kechuanan capsule could apparently prolong the coughing incubation period and reduce coughing frequency, and could apparently increase phenol red in mice pulmonic excretion. The average area and integral optical density of NF-κB in asthma mice bronchus wall was excess than the nomal group and all of the Kechuanan capsule groups remarkably (p〈0.01). Conclusion: Kechuanan capsule has significantly antitussive and expectorant effects, and can reduce the expression of NF-κB of asthma mice lung. This may be the possible mechanism.
