摘要
【目的】了解植物次生生长调控网络以及挖掘新型次生生长相关NAC转录因子。【方法】通过生物信息学手段对19个拟南芥次生生长相关NAC转录因子进行氨基酸序列比对及系统发生树的构建,寻找保守结构域,并对其保守结构域及蛋白质的二级结构、亚细胞定位等进行分析、预测。【结果】通过对19个拟南芥NAC转录因子的N端保守域进行划分,共发现A、B、C、D、E5个保守性不同、同时含有多个化学修饰位点以及疏水性区域的亚结构域,其中亚域D可能涉及DNA绑定及核定位信号功能,而位于多变氨基酸序列C端的F区与G区,可能涉及转录激活功能,而该区域也是划分次生生长相关NAC转录因子亚家族的重要区域。【结论】拟南芥NAC类转录因子的保守结构域,对于次生生长NAC转录因子家族功能的发挥具有重要作用。
[Objective] The study was done to better understand the control network of plant secondary growth and find new NAC transcription factors with secondary growth in Arabidopsis. [Method] Multiple alignment was done and phylogenetic tree was built to find the conservative domains of NAC transcription factors with secondary growth in Arabidopsis. Analysis and forecast of these conservative domains as well as secondary structure were done with the help of bioinformational tools. [Result] There were five subdomains in N side of amino acid which were A, B, C, D and E. These subdomains contained many chemical modification sites and hydrophobicity areas, and the conservative characters of which were different. Subdomain D was putative to bind DNA and function as nuclear localization signal, F area and G area in C side were invovled in transcription activation and identify the NAC transcription factor subfamilies with secondary growth. [Conclusion] The conservative domains of NAC transcription factor with secondary growth in Arabidopsis play an important part in the function of NAC transcription factor family with secondary growth.
