摘要
实验用ST.ThomasⅠ号液和改良灌注猪心并保存于4℃该液中,观察其对不同保存时间心血管形态学主为化。方法分别于灌注后妈了时、4h、6h、7h和保丰了4h、5h后离体心脏再灌注复苏30min-52min后,行冠脉墨汁灌注、取心室肌组织学切片,电脑图象分析心肌微血管蜜度、观察其超微结构。
Objective To study the effect of potassium on porcine ischemic myocardium and microvasculature.Methods Porcine hearts were flushed and immersed by 4°C St.Thomas No.1 solution (Group 1,n=6,k + 20mmol/L) and by modified Collins solution ( Group 2, n=6, k + 115mmol/L).After preserval for 4~7 h, all were performed coronary ink perfusion and four of the isolated hearts (two in each group) were reperfused and resuscitated. The morphological changes of myocardium and microvasculature were studied.Results 1.The microvascular density in group 1 was higher than that in group 2 (P<0.001).At the end of preservation, the edema was more severe in group 1 than in group 2.There was no obvious difference in myocardial ultrastructures, nuclear chromatin and mitochondral edema.However,after reperfusion and resuscitation, the edema and scarcity of glucose granules were more severe in group 2 than in group 1.Conclusion Extremely hyper potassium (k + 115 mmol/L ) is more harmful to myocardial and endothelial ultrastructure.
