摘要 介绍一种用四氦唑蓝为底物的酶反应定量比色分析法。方法用MTT法测定了丙烯酰胺对体外培养的大鼠胚胎中脑神经细胞的毒性。结论MTT法用于测定细胞毒性作用具有快速,正确等优点。 Objective To introduce a quantitative colorimetric assay based on a tetrazolium salt (MTT). Methods To testify the toxic effect of acrylamide on neurons from rat embryo midbrain in vitro by using MTT. Results The absorbance was directly proportional to the number of cells and the linearity was good in the range from 1×10 4 to 4×10 5 cells/ml culture fluid;Acrylamide was proved to be a cytotoxicant for neuron and its IC 50 value was 85 ug/mg.Conclusion The method of MTT can be used to measure neuron cytotoxicity.The main advantages of this assay are its rapidity and precision.
