摘要
为获得高质量的冷季型草坪草基因组DNA,用改良和优化常规CTAB提取方法,提取冷季型草坪草基因组DNA,并对所得DNA的质量和浓度进行了检测。结果表明:改良CTAB-B法要好于常规CTAB法和改良CTAB-A法,提取的基因组DNA的OD260/OD280在1.8~1.9之间,电泳条带清晰,RNA消化彻底,DNA无明显降解,其含量和纯度均能满足基因工程操作的要求,且酶切效果理想。
Genome DNA was extracted by the improved CTAB method from Cool-season turf grass and the quality and concentration of DNA were detected. The results indicated that improved CTAB-B method was a simple and high efficient method which could obtain the high quality of genome DNA , the value of OD260/OD280 was 1.8 to 1. 9, the RNA was eliminated completely without obvious degradation of DNA, the content and purity can meet the requirements of genetic engineering, and digested the results were satisfactory.
