神经节苷脂GM1在BMSCs向神经细胞分化的作用

摘要 目的研究体外使用神经节苷脂GM1诱导骨髓间充质干细胞(BMSCs)向神经细胞分化的作用。方法从健康志愿者髂骨处抽取骨髓5mL,体外分离、培养、扩增后,用含有10μg/mL、60μg/mL、90μg/mLGM1的无血清培养基诱导BMSCs向神经细胞方向分化。0.5h、3h、6h、24h后使用倒置显微镜和免疫细胞化学方法观察诱导前后细胞的变化。结果诱导6h后各组均有部分细胞胞体收缩、伸长突起,表现出神经细胞的形态。0.5h、3hNF、MAP-2阳性细胞比例无统计学意义(P>0.05),6h、24hNF、MAP-2阳性细胞比例无统计学意义(P>0.05),6h、24h分化率高于0.5h和3h(P<0.05),含60μg/mLGM1的无血清培养基诱导后细胞的分化率高于10μg/mL组和90μg/mL组。结论含60μg/mLGM1的无血清培养基可以有效地诱导BMSCs向神经细胞分化。
