摘要
目的探讨君子兰生物总碱(TACM)对大鼠肝脏、肾脏组织功能和抗氧化能力的影响,用以评价药物的安全性和对肝肾组织的保护作用。方法按低、中、高三个剂量组,经灌胃给予实验Wister大鼠TACM4个月,测定尿糖含量、尿蛋白水平,测定血浆肝脏及肾组织功能相关指标,测定肝肾组织丙二醛(MDA)的含量、超氧化物歧化酶(SOD)活性、谷光甘肽过氧化物酶(GSH-Px)活性,同时留取肝。结果实验组大鼠与对照组比较各组尿糖、尿蛋白、肝肾功能相关指标无显著差异(P>0.05);MDA无明显变化(P>0.05)、SOD和GSH-Px活性在长期大量应用组显著增强(P<0.05),且有一定的量效依赖关系。结论TACM具有使肝肾组织抗氧化能力增强、降低氧化损伤的作用,对大鼠肝脏、肾脏未见慢性毒性作用,提示应用TACM对肝肾组织可有不同程度保护作用。
Objective To investigate the effects of total alkaloid of clivia miniata(TACM)on the hepatorenal function and antioxidant ability, and evaluate the reliability and protective effects of this drug on liver and kidney in rats. Methods TACM with low, middle and high dose was taken orally by three groups of experimental Wister rats for 4 months. MDA content and the activity of SOD and GSH-Px in liver and kidney of rats were determined. Results There was no significant difference between the urine glucose content, urine protein content as well as the relative function marker of liver and, kidney of rats in experimental group compared with that of in control group(P 〉 0. 05 ), MDA content also showed no significant change; The activities of SOD and GSH-Px were greatly increased in the high dose group (P 〈0.05 ), which showed a dose-effect dependent appearance. Conclusion TACM could enhance the antioxidant ability and reduce the antioxidant stress injury of liver and kidney in rats without toxic effects. Our findings suggest that the application of TACM could produce protective effects on liver and kidney in rats.
