摘要
以急性早幼粒白血病人短型融合蛋白的PML与RARα结合部位的13个氨基酸序列制备合成肽,交联甲状腺球蛋白免疫Balb/c小鼠,通过杂交瘤技术获得两株单抗,经鉴定与相关蛋白无交叉反应,说明此融合部位外露,可以制备出针对性抗体。
A peptide of 13 amino acids correspoding to the short type PML-RAR a juntions was synthesized as an antigen. Peptide was coupled with thyroglobulin by MBS. By using the hybrido-ma techique we have generated two mouse monoclonal antibodies against short type PML-RARa juntion point. The isotype of both the monoclonal antibodies was IgG1 and has no cross-reaction with related protein , indicating the specificity for PML-RAR a S type fusion-point epitope.
