摘要
采用酶免疫和放射免疫法分析了再生障碍性贫血(AA)患者外周血T淋巴细胞亚群、血清肿瘤坏死因子(TNF)、γ-干扰素(IFN-γ)和IL-2水平,以及患者外周血单个核细胞体外诱生的上述细胞因子水平。结果表明:患者CD4/CD8细胞比值降低或倒置(P<0.001),血清TNF和IFN-γ水平显著增高(P<0.01),外周血单个核细胞诱生的TNF、IFN-γ和IL-2水平均明显高于正常对照(P<0.01)。提示T淋巴细胞亚群比例失调和TNF、IFN-γ等造血负调控因子的过量分泌可能与AA的发病机制有关。
The aim of this study was to investigate the immunopathogenesis of aplastic anemia ( AA). T cell subsets and the levels of tumor necrosis factor (TNF), interferon gamma (IFN-7), interleukin-2 (IL-2) both in the serum of 40 patients and in the culture supernatant of peripheral blood mononuclear cells (PBMNC) in the presence of phytohemagglutinin (PH A) of 10 patients with AA were analysed using enzyme immunoassay and radioimmunoassay methods. The results showed that the ratio of T cell subsets CD4/CD8 significantly decreased or reversed (P<0. 001). The levels of TNF and IFN both in the serum and in the culture supernatant of PBMNC significantly increased than normal control (P<0. 01). IL-2 in the serum was no difference with normal control (F>0. 05) but significantly increased in the culture supernatant of PBMNC (P<0. 01). The proportional imbalance of T cell subsets and overproduction of hematopoietic inhibitory cy-tokines TNF and IFN may play an important role in the progression of AA.
