摘要 棉属40个不同种、远缘杂种的宿根棉株的茎尖。腋芽离体培养结果如下:在0.1mg/1ZT,0.02mg/1IBA,1.9g/1KNO_3,3%蔗糖的MS培养基上,诱导茎尖和腋芽为幼芽。幼芽诱导率达87.5%;幼芽增殖与基因型关系密切;生根诱导依赖于幼枝的生长特征,KT和IAA的配比及其含量。壮苗生长是提高试管苗移栽成活率和提高试管苗应用效果的主因。 Culture of shoot apexes and axillary buds of cotton in vitro had resulted in the regeneration of a number of plantlets of dwarf type, but few instances of seedling survival after transplanting had been reported. The present research was aimed at developing whole plants from shoot tip culture, enhancing the rate of post-transplant survival and discussing the potential use in germplasm research. Experiments were carried out with 40 specific materials,including cultivated species, wild species and distant hybrids in Gossypium. Qn MS medium supplemented with 0.1 mg/1 ZT, 0.02 mg/1 IBA, 1.9 g/1 KNOs and 3 % sucrose, the rate of budlet induction was 87.5%. The effect of genotype on budlet propagation was obvious. Rooting and vigorous growth of plantlets were a key to transplanting success. Clo nal plants and their progenies were normal in growth and fertility.
