复方地黄对早老性痴呆小鼠形态影响和相关增殖因素变化的研究

摘要 目的研究复方地黄对早老性痴呆小鼠的神经生长因子改变及形态学的影响,进一步探讨复方地黄对老年痴呆的作用机制。方法将实验小鼠随机分为四组,实验组给予复方地黄混浊液2ml/d灌胃。治疗组给予安理申混悬液400 mg/kg.d,空白对照组和模型对照组用等量生理盐水。3周后,各组小鼠进行行为学测试,应用免疫组化法检测脑组织神经生长因子改变及电镜观察形态学改变。结果复方地黄能够改善SAM-P/8小鼠的学习和记忆能力,减少痴呆模型小鼠的包涵体数量,增加大脑皮层及海马区神经营养因子的含量,对SAM-P/8小鼠具有较好防治作用。结论复方地黄能够明显改善早老性痴呆小鼠的学习能力和蛋白表达。 Objective To investigate the effect of compound rehmannia on change of nerve growth factor(NGF) and apoptosis--associated gene in hippocampal area of SAM--P/8 rats. Methods Experimental rats were randomly divided into 4 groups. Experimental group was lavaged by compound rehmannia (2ml/day, individual). Negative group was lavaged by Donepezil Hydrochloride (400mg/kg,individual) ,and normal group and model contrast group was lavaged by equal Sodium Chloride. We observed every group behavior, and investigate the change of nerve growth factor (NGF) and changes of its biological characteristics were determined using electron microscopy cell. Results Compound rehmannia could enhance the SAM--P/8 rats study and memory ability, strengthen the nerve growth factors (NGF) volume. Conclusions Compound rehmannia have better effect on SAM--P/8 rats.
