摘要
[目的]建立大樱桃砧木吉塞拉组培快繁技术体系。[方法]以吉塞拉5号的茎尖分生组织为外植体,从材料丛生芽的诱导、继代、生根以及炼苗与移栽方面研究其组培快繁技术。[结果]结果表明,适宜丛生芽诱导的培养基为MS+6-BA 1.00 mg/L+NAA 0.03mg/L+KT 1.50 mg/L+GA30.20 mg/L;试管苗在以1/2 MS+IBA 1.50 mg/L的培养基上生根效果好;以1/2草粪+1/2园土为移栽基质时,小苗成活率高。[结论]为大规模、快速生产吉塞拉组培苗提供理论和实践依据。
[Objective] The research aimed to establish the tissue culture and rapid propagation technology system of big sweet cherry rootstock Gisela.[Method]With the shoot apical meristem of Gisela No.5 as material,the paper studied the tissue culture and rapid propagation technology from axillary buds induction,subculture,rooting and plantlet hardening and transplantation.[Result]The results showed that the optimum medium for axillary buds induction was MS+6-BA 1.00 mg/L +NAA 0.03 mg/L +KT 1.50 mg/L +GA3 0.20 mg/L;1/2 MS+IBA 1.50 mg/L was suitable for the test tube plantlet rooting.The young plantlet had high survival rate when took the 1/2 straw dung and 1/2 garden soil as transplantation substrate.[Conclusion]This study will provide the theoretical and practical basis for rapid production of tissue culture of Gisela No.5.
