摘要
[目的]为猪异种器官移植的免疫识别机理的研究奠定基础。[方法]采用RT-PCR方法扩增五指山猪DQA基因cDNA,然后进行测序和序列分析。[结果]海南五指山猪SLA-DQA基因序列长度为768个核苷酸,其中1~765为完全阅读框编码255个氨基酸残基。五指山猪SLA-DQA基因cDNA序列及其推导的氨基酸序列与我国小型猪同源性最高,与人相应基因的同源性明显高于与小鼠的同源性。[结论]将五指山猪作为适合于进行异种器官移植的中国猪种,对免疫遗传学特性系列研究具有重要意义。
[Objective] The study aimed to lay a foundation for studying immune recognition mechanism in xenotransplantation.[Method] SLA-DQA gene of Hainan Wuzhishan miniature pig was cloned by RT-PCR and sequenced.[Result] The SLA-DQA gene sequence of Wuzhishan miniature pig contained 768 nucleotides,among which 1-765 was the complete open reading frame,encoding 255 amino acids.SLA-DQA cDNA sequence and its deduced amino acid sequence of Wuzhishan miniature pig were most similar to those of the other miniature pigs in China.When compared with corresponding cDNA sequences and the amino acid sequences in other species,the homology with human was obviously higher than that with mouse.[Conclusion] As the Chinese pig breed suitable for xenotransplantation,Wuzhishan miniature pig has significant importance to serial studies on immunogenetics features.
