摘要
报道一种简易改良骨髓活检块GMA塑料包埋法,结合冷固定、冷脱水、冷浸渍和冷包埋,于4℃真空下聚合的有效制冷系统,不仅容许缓慢聚合而又使许多细胞抗原因聚合过程中过度产热所致热损伤的防止而得以保留。为研究同一切片内骨髓细胞形态和免疫组化分析提供了一种有效诊断工具。CML患者切片以CD八h单抗染色后于不同成熟阶段之巨核细胞之胞浆均呈强阳性反应,方法简单而无需昂贵设备,适于以取得之小块骨髓在任何血液实验室中常规进行。
In the study, a simple improved procedure allows bone marrow biopsy cores to be plastic embedment in glycol methacrylate (GMA), coupled with fixed in cold Bouin's solutfion, dehydrated in cold methanol infiltrated and embedded in cold GMA, then polymerized at 4 ℃ under vacuum provided an effective cooling system which permites slow polymerization, but also preserves a wide rang of cellular antigens by preventing heat damage due to excess 'rise of temperature- It was a useful diagnostic tool to study of the morphology and immunohistochemical, analysls of bone marrow cells in the same section. The section was stained for antibody to CD41antigerl showing strong positive reaction throughout cytoplasm of megakaryocytes in different stages of maturation from patients of chronic myelold leukemia. The method is simple, requires no expensive equipment and is suitale for routine process of small bone marrow cores in any hematology laboratory.
