摘要
目的观察超微六味地黄汤对被动吸烟小鼠胚胎体脑发育及胎脑生长激素(GH)、生长抑素(SS)的影响,反证中医"肾"与"脑"的关系。方法采用孕鼠被动吸烟造模,动物随机分为正常组(A)、模型组(B)、传统组(C)、超微1/3组(D)、超微全量组(E)5组,孕19(E19)天处死母鼠,称取体重后迅速剥离胎脑称重匀浆,采用酶联免疫吸附(ELISA)法测定胎脑匀浆液GH、SS水平。结果被动吸烟可使胎仔发育迟缓,影响胎鼠脑组织GH、SS水平,与正常组比较差异有显著性(P<0.05);各治疗组能增加胎鼠体、脑质量,调节紊乱的GH、SS水平,一定程度改善胎脑发育(P<0.05或P<0.01),且超微全量组在改善胎鼠体脑重及体脑比重方面优于传统组或超微1/3组(P<0.05或P<0.01)。结论超微六味地黄汤可能通过神经内分泌机制显著促进宫内发育迟缓(IUGR)小鼠胎仔的体、脑发育,超微1/3量与传统组之疗效相当,提示"肾"通于"脑"理论的合理性。
Objective To observe the effect of Ultra - micro - LiuwciDihuang Decoction on the embryo brain development and the influence on brain growth hormone(GH), Somatostatin(SS) in prengnant mice of passive smoking, then to demonstrate the relationship between brain and kidney in TCM. Methods Animals were divided into 5 groups at random: normal group(A) , model group(B) , traditional Liuwci Dihuang Decoction group (C) , 1/3 dosage of Ultra- micro -LiuweiDihuang Decoction group(D) and all dosage of Ultra - micro -LiuweiDihuang Decoction group(E). The IUGR model was established by passive smoking. On the 19th day of gestation, all mice were anatomized to scale the body and brain weight of lively embryos,then a part of brain tissue was homogenated. The levels of GH and SS in brain tissues were measured by ELISA. Results Passive smoking could decrease the body and brain weight, influence the level of GH and SS in brain tissues. Compared with A and B groups, the C, D and E groups could increase body and brain weight, regulate the level of GH and SS, improve the brain development of IUGR mice obviously(P 〈 0.05 or P 〈 0.01 ). Moreover, there was significantly difference among E and C group or D group in body and brain weight(P 〈 0.05 or P 〈 0.01 ). Conclusion Ultra - micro - Liuwei Di- Huang decoction can improve the body and brain development of IUGR mice obviously by neuroendocrine mechanisms. 1/3 dosage of Ultra -micro- LiuweiDihuang Decoction has the same curative effect as traditional Liuwei Dihuang Decoction, Which proves the theory that "kidney communicated brain"in TCM is reasonable.
