CO2气腹对乙酰肝素酶、血管内皮生长因子在裸鼠胃癌组织中表达的影响

摘要 目的通过建立人胃癌裸鼠腹腔种植瘤模型,探讨CO2气腹对胃癌组织中乙酰肝素酶(HPA)、血管内皮生长因子(VEGF)表达的影响及意义。方法27只裸鼠分3组:对照组、5mmHgCO2气腹组及8mmHgCO2气腹组,每组9只。人胃癌细胞SGC-7901裸鼠腹腔注射后建立CO2气腹胃癌细胞裸鼠腹腔种植模型,气腹持续1h。处理后4周处死解剖裸鼠,记录腹腔各脏器、穿刺口和腹壁切口成瘤情况和肿瘤数,测量肿瘤最大直径、总质量,并检测胃癌种植瘤组织中HPAmRNA和蛋白表达及VEGF蛋白的表达。结果各组裸鼠腹腔种植瘤结节数、平均最大直径、质量相比无统计学差异(P>0.05)。各组裸鼠胃癌组织中HPA和VEGF的表达水平相比无统计学差异(P>0.05)。结论5mmHg和8mmHgCO2气腹对胃癌细胞裸鼠腹腔内增殖和侵袭转移无明显影响,也未明显促进种植瘤组织中HPA和VEGF的表达。 Objective To observe the direct influence of CO2 pnemumoperitoneum on the growth and metastasis of seeded SGC-7901 cells in nude mice, and to investigate its effect of CO2 pnemumoperitoneum on the expressions of heparanase (HPA) and vascular endothelial growth factor (VEGF) in the tissue of tumor. Methods Totally 27 nude mice were randomly divided into 3 groups (9 in each group ) , 2 groups of CO2 pnemumoperitoneum (5 or 8 mmHg CO2) and control group. After SGC-7901 cells suspension (2 × 10^6) were injected into peritoneal cavity of all nude mice, the CO2 pnemumoperitoneum pressure at 5 or 8 mmHg was established for 1 h. The control group also underwent a midline incision, which was closed 1 h later as in CO2 pnemumoperitoneum groups. Four weeks later, all nude mice were killed, then the tumorigenesis, the total weight, mean max diameters of tumor mass in the peritoneal cavity were observed and recorded. The expressions of HPA and VEGF in the xenograft were measured by immunohistochemieal method. Results There was no significant difference in the number of abdominal cavity nodules, the total weight and the mean max diameters of each group (P 〉 0. 05 ). The expression of the HPA and the in gastric cancer tissues showed no significantly difference in each subgroup (P 〉 0. 05). Conclusion CO2 pnemumoperitoneum does not accelerate the the growth and invasion of SGC-7901 cells in vivo, and the expression of the HPA and the VEGF in the tumor mass.
