摘要
目的观察姜黄素对膀胱肿瘤T24细胞中胰岛素样生长因子-1(IGF-1)表达的影响。方法用不同浓度的姜黄素作用于膀胱肿瘤细胞株T24,用MTT法测定细胞抑制率,用流式细胞仪测定细胞凋亡结果,用RT-PCR测定不同姜黄素浓度对肿瘤细胞株胰岛素样生长因子-1(IGF-1)及胰岛素样生长因子-1受体(IGF-1R)mRNA表达量的影响。结果姜黄素对膀胱肿瘤细胞株的作用具有时间和剂量依从性,姜黄素能够诱导膀胱肿瘤细胞株凋亡,姜黄素可使肿瘤细胞株的IGF-1mRNA表达量降低。结论姜黄素可能会通过胰岛素样生长因子-1(IGF-1)通路抑制膀胱肿瘤T24细胞的生长,促进其凋亡。
Objective To observe the effect of curcumin on insulin-like growth factor-1 action in T24 human bladder tumor ceils. Methods The bladder tumor cell lines were administered differ concentration of curcumin. The rate of cell inhibition,the rate of apoptosis and express of IGF-1 and IGF-1R mRNA were determined with MTT,flow cytometer and RT-PCR,respectively. Results Either the rate of cell inhibition or the rates of apoptosis depend on the curcumin dose and the action time of curcumin. The quantity IGF-1 mRNA was reduced when the curcumin doses increased. Conclusion The curcumin can depress the growth and enhance the apoptosis on insulin-like growth factor-1 action in T24 human bladder tumor cells.
