流式微球技术检测参芪扶正注射液对化疗小鼠Th1/Th2平衡状态的影响 被引量:4

Determination of the Effect of Shenqi Fuzheng Injection on the Balance of Th1/Th2 by Cytometric Bead Array in S 180-bearing Mice with Chemotherapy

摘要 目的:研究参芪扶正注射液对化疗小鼠Th1/Th2平衡状态的影响。方法:57只荷瘤小鼠随机分成5组,分别为荷瘤对照组、环磷酰胺组、参芪扶正注射液高剂量组(8g·kg-1组)、临床等效剂量组(4g·kg-1组)和低剂量组(2g·kg-1组)。后4组给予环磷酰胺ip2次造模。荷瘤对照组及环磷酰胺组ip生理盐水,1次/d,其他3组ip相应剂量参芪扶正注射液。9d后摘小鼠眼球取血约0.5mL进行流式微球(CBA)检测,再脱颈处死小鼠,称取肿瘤重量,计算肿瘤系数。结果:参芪扶正注射液临床等效剂量组(4g·kg-1)可显著降低化疗小鼠的肿瘤重量和肿瘤系数(P<0.01),提高化疗小鼠的TNF-α及IL-2的浓度(P<0.01~0.05)。结论:参芪扶正注射液4g·kg-1可诱导化疗小鼠的Th1/Th2平衡向Th1细胞占优势的方向转化。 Objective:To study the effect of Shenqi Fuzheng Injection on the balance of Th1/Th2 in S180-bearing mice with chemotherapy. Methods: 45 of 57 tumor-bearing mice were given cyclophosphamide twice by intraperitoneal injection, and then randomly divided into four groups: the model group, the high dose group (8 g·kg^-1), the clinical equivalent dose group(4 g·kg^-1) and the low dose group (2g·kg^-1) of Shenqi Fuzheng Injection. The model group and the 12 tumor-bearing mice without cyclophosphamide injection were given physiological saline by intraperitoneal injection once a day, while the others were given Shenqi Fuzheng Injection in the same way. 9 days later,the eyes and 0.5 mL blood were obtained before each mouse was sacrificed. The blood was for cytometric bead aray(CAB)test. The tumors were weighed, and the tumor index was calculated. Results: Shenqi Fuzheng Injection, in the clinical equivalent dose, could significantly depress the weights of tumors and the tumor indexes( P 〈 0.01 ), and elevate the contents of TNF-α and IL- 2 (P 〈 0.01 -0.05) in S180-bearing mice with chemotherapy. Conclusion: In clinical equivalent dose(4 g·kg^-1), Shenqi Fuzheng Injection could make Thl cells to be predominant to Th2 ceils in S180-bearing Mice with chemotherapy.
