摘要
目的:探讨头部电针透穴疗法治疗帕金森病的作用机制。方法:将36只Wistar大鼠随机分为正常组、假手术组、模型组、透针组,以6-羟基多巴胺(6-OHDA)左侧纹状体注射法制备偏侧帕金森病大鼠旋转模型,透针组穴取"百会"透"太阳",每日1次,6日为一疗程。其他3组不予治疗,观察2个疗程。采用以HE染色观察大鼠黑质神经细胞组织形态变化,以RT-PCR技术检测酪氨酸羟化酶(TH)及多巴胺转运体(DAT)mRNA的表达。结果:透针组TH mRNA(1.22±0.19)及DAT mRNA(0.62±0.11)均较模型组TH mRNA(0.65±0.17)及DAT mRNA(0.41±0.08)表达明显增加(均P<0.05)。大鼠黑质神经细胞透针组较模型组数量增多,细胞变性减轻。结论:头部电针透穴疗法能使帕金森病模型大鼠黑质TH及DAT mRNA表达增高,进而可以促进多巴胺的合成与重摄取,以达到治疗帕金森病的目的。
Objective To explore the mechanism of eleetroacupuncture scalp point-through-point therapy in the treatment of Parkinson's disease. Methods Thirty-six Wistar rats were randomly divided into a normal group, a sham-operation group, a model group and a point-through-point therapy group. 6-OHDA was injected into left striaturn to made lateralization Parkinson's disease rat model. The point-through-point therapy group was treated with electroacupuncture at "Baihui" (GV 20)-through-"Taiyang" ( EX-NH 5 ), once each day, 6 days constituting one course, for 2 courses, and the rats of other groups were not treated. HE staining method was used for observation of the histo-morphologic changes of the substantia nigra neurons, and RT-PCR for the expression of tyrosine hydroxylase (TH) and dopamine transporter(DAT) mRNAs. Results The expressions of TH mRNA (1.22 ± 0.19) and DAT mRNA (0.62±0.11) in the point-through-point therapy group were significantly higher than (0. 65± 0.17) and (0.41±0.08) in the model group, respectively (all P〈0.05). As compared with the model group, the number of neurons in the substantia nigra increased and degeneration of the neurons relieved in the point-through- point therapy group. Conclusion The electroacupuncture scalp point-through-point therapy can increase expressions of TH mRNA and DAT mRNA in the substantia nigra in the Parkinson's disease model rat, and promote synthesis and reuptake of dopamine, hence Parkinson's disease is cured.
