摘要
目的建立HPLC-ELSD法测定胆酸止咳片中猪去氧胆酸的含量。方法采用Eclipse XDB-C18色谱柱(4.6×250mm,5μm),流动相为甲醇-0.1%甲酸(75∶25),柱温30℃,流速1.0mL.min-1,检测器为ELSD,漂移管温度为74℃,氮气流速为2.1L.min-1。结果本法猪去氧胆酸的线性范围分别为1.056μg～5.280μg(r=0.9997),平均回收率(n=5)为97.41%,RSD=0.9%。结论方法简便,结果准确,重现性好,可用于胆酸止咳片的质量控制。
OBJECTIVE To establish a HPLC-ELSD method for the hyodeoxycholic acid quantitative analysis of Danshuan Zhike tablets. METHODS Chromatographic condition included Eclipse XDB-C18 （4.6 × 250mm, 5μm）and mobile phase was methanol-0.1% formic acid solution（75 ： 25）, the flow rate was 1.0mL· min-1, the column temperature was 30℃, the detector was ELSD, the drift tube temperature was 74℃ and the flow rate of N2 was 2.1L· min-1. RESULTS The method was linear within the range of 1.056μg--5.280μg, its recovery was 97.41%, RSD was 0.9 %. CONCLUSION The method was accurative, reproducible and can be used to con- trol the quality of Danshuan Zhike tablets.
