茶多酚抑制卷烟烟气对支气管上皮细胞氧化损伤的作用 被引量:6

摘要 目的观察茶多酚抑制卷烟烟气诱发支气管上皮细胞(BEAS-2B)氧化损伤的作用。方法细胞培养液中加入卷烟烟气吸收液造成损伤模型,并在培养液中加入茶多酚进行保护,通过检测细胞外过氧化氢、超氧阴离子和羟自由基的变化和乳酸脱氢酶的溢出观察细胞氧化损伤的情况。结果细胞经卷烟烟气染毒后,细胞内和细胞外自由基含量显著增高且呈剂量效应关系;且引起细胞内的乳酸脱氢酶溢出,茶多酚对卷烟烟气诱发的活性氧(ROS)增高及乳酸脱氢酶(LDH)溢出有明显的抑制作用。结论卷烟烟气能造成细胞的氧化损伤,茶多酚对其氧化损伤具有抑制作用。 Objective To observe oxidative damage in human bronchial epithelial cells ( BEAS -2B) induced by cigarette smoke condensate ( CSC ) and protection effect of tea polyphenols. Methods To make damage model, we added CSC in culture fluid, and added tea polyphenols for protection. The observation of oxidative damage was based on the measurement of superoxide anions( O2 ^- ) ,hydroxyl radicals ( · OH)and hydrogen peroxides (H2O2 ). LDH was measured by AUTOLAB. Results The reactive oxygen species ( ROS ) production including H2O2, O2 ^- and · OH increased remarkably which induced by CSC in BEAS -2B cells. These changes could be inhibited effectively by tea polyphenols. Conclusion CSC causes oxidative damage to BEAS- 2B cells, which can be protected by tea polypbenols
