APAAQ分光光度法测定血清微量铜的研究 被引量:1

摘要 参照文献报道合成新双偶氮显色剂5-[(偶氮本基)偶氮)-8-氨基喹啉(APAAQ),研究其与血清铜离子的显色条件,并首次将其应用于血清铜的测定,获得满意效果。研究表明:在混合表面活性剂SDBS+OP存在下,弱酸性介质中(pH4.0~7.0),APAAQ能选择性地与铜离子发生灵敏的显色反应,形成1:3的紫色络合物,常温下稳定4h.λmax为550nm,摩尔吸光因数K550=3.24×104cm2/mol,精密度为2.35%,平均回收率为101.82%,血清铜在0~28μmol/L范围内符合Lambert-Beer定律,大多数金属郭子不影响测定。该法(Y)是目前测定血清铜的最理想分光光度法之一,可与AAS法(X)相炮美,P>0.05,直线回归方程为Y=0.9862X+0.0923,r=0.9920,P<0.001,适于临床推广。 Having synthesized a new diazo-chromogenic compound 5-[(azophenyl)-zoo]-9-aminoquinoline(APAAQ) and we studied its optimum reaction condition with serum divalent coppers and primarily applied it to de-termine serum divalent coppers with satisfactory results. The results was showed as follows: in a weak acid medi-um of pH 4. 0~7.0,divalent coppers reacts with APAAQ selectively to form a 1 : 3 purple complex compoundsensitively,which can be stable for 4 hours at ambient temperature, in the presence of mixed surfactant SDBS&OP- The molar absorptivity of Cu-chelate was found 3. 24× 104/cm2/mol correspondingly at (max = 550 nm.The precision and the average recovery are 2. 35% (n = 20) and 101. 82 % (n = 6) respectively. Beer's concentra-tion law is obeyed in the concentration range of 0~ 28μmol/L of divalent coppers. Large amounts of serum usualmetal ions are tolerated for the determination. The method is one of the idealist spectrophtrophtometric methodsof determination of serum divalent coppers. The result comparison of this method (Y) and flame atomic absorptionspectrometry (AAS) (X) showed that: there were no significant differences between them(P>0. 05), linear re-gression equation was Y= 0. 986 2X+ 0.092 3,r= 0. 992 0, P<0. 001. So this method was suitable to be general-ized in clinical medical laboratory.
