摘要
目的:将传代的人牙髓细胞培养于加入10%FCS的DMEM液中,其中含50μg/mgL-VitC、10mmol/Lβ-磷酸甘油钠,观察牛血浆纤维粘连蛋白(FN,20μg/ml)对细胞的增殖、表型的影响。方法:细胞培养,免疫组化染色,扫描电镜。结果:FN(20μg/ml)在细胞增殖期可促进细胞增殖,但在静止期则无明显的促增殖作用,也不能改变细胞的形态、生长方式;两组细胞的ConA、WGA凝集素受体的表达也无差异。扫描电镜证实:两组细胞表面光滑。结论:FN(20μg/ml)可促进含10%FCS的DMEM液中的人牙髓细胞的增殖,但不影响其表型。
Aim: To evaluate the effects of fibronectin (FN,20μg/ml)on cultured human dental pulp cells that incubated in DMEM supplemented with 10% FCS, 50μg/ml acorbic acid and 10mmol/L β-sodium glycero phosphate, the proliferation or phenotype of cells were observed. Methods: Cell cultured. SEM. Immunohistochemistry staining. Results: Fn(20μg/ml) stimulated the cells proliferation in proliferating stage and had no effects in stationary stage of culture, FN(20μg/ml) didn't change the morphology and growth stale of the cells. There was no significant difference in expression of ConA, WGA receptors between two groups. Scanning electron microscopy visualization of the fibroblasts revealed the characteristic smooth surface of cells. Conclusions: FN(20μg/ml) stimulated the proliferation of human dental pulp cells in DMEM supplemented with 10% FCS, but there was no effects on the phenotype of cells.
