摘要
使用c-fos原癌基因表达产物免疫组化标记方法,观察了安宫牛黄丸对中枢神经元的活化作用。结果显示:1.该药能够活化包括脑干、丘脑及皮层的神经元,证实了前人关于安宫牛黄丸可能通过激活脑干网状上行激活系统而达到促进清醒作用的推测;2.发现在下丘脑内有相当数量的神经原被标记;提示安宫牛黄丸可能通过激活神经内分泌系统及与情绪、意识等有关的下丘脑等中枢起作用;3.杏仁核、膈核、终纹床核等部位大量神经元被活化,说明安宫牛黄丸可能通过激活边缘系统而发挥作用。4.广泛的大脑皮层神经原被活化,提示安宫牛黄丸对皮层神经原可能还有直接的作用。在对照组大鼠脑内仅见稀疏散在c-fos免疫阳性神经元分布。
This study was conducted using immunohistochemistry to detect the expression of the prooncogen cfos in the brain of the rats fed with Angongniuhuang pill.An antiserum against Fos oncoprotein was used to immunohistochemically detect the activated cells in the brain.In the experimental group,a great amount of cFosimmunoreactive(ir) nuclei were identified to be present in the brain stem as well as thalamus,hypothalamus,limbic system and cortex.In the brain stem,many labeled nuclei were seen in the central gray substance,geniculate body,the superior colliculus,the dorsal inferior colliculus,the nncleus of solitary tract,the interpeduncular nucleus,the pontile nuclei,the medium reticular nucleus and lateral magnocellular reticular nucleus,In the thalamus,the activated neurons were concentrated in the paraventricular thalamus nucleus and the nucleus of medial geniculate body.These results were identified with the theory that the pill activated on the neurons system through the ascending reticular activating system.However,our results showed the activated neurons in more wider areas including the hypothalamus,the limbic system,the cortex et al.These date explained that this pill have more complex effects on the central system. Only were a few labeled nuclei seen in the control group rat.
