摘要
为了探讨扶正化瘀方抗肝纤维化的作用机制。以体外培养的NIH/3T3细胞作为肝脏成纤维细胞的替代模型,用扶正化瘀方药物血清,以10%的浓度被添加到培养液中,通过[^3H]TdR掺入法和免疫细胞化学,观察药物血清对3T3细胞[^3H]-TdR掺入量和胶原生成的影响。结果表明药物血清组[^3H]-TdR掺入量明显少于正常大鼠血清组。药物血清组Ⅰ、Ⅲ、Ⅳ型胶原量均明显低于正常大鼠血清组,其中Ⅰ。
In order to search the mechanism of FZHYF' s inhibiting the progress of liver fibrosis. By method of [3H] - TdR incorporation and immunocytochemistry we observed the effects of the FZHYF - treated rat sera, which were obtained from the rats administrated orally by FZHYF, on the proliferation and collagen synthesis of NIH/3T3 cell which is a putative model in vitro instead of fibrobalsts in liver in vivo. The re- suits showed that FZHYF - treated rat sera could significantly inhibit [3H] - TdR incorporation of N1H/ 3T3 cell. The FZHYF -treated rat sera could significantly decreased the collagen ( I -, UK IV ) synthesis of 3T3 in vitro. Particularly the collagen( I > HI )were obviously decreased. These indicated that FZHYF - treated rat sera significantly inhibited proliferation >and collagen synthesis of NIH/3T3 cell in vitro.
