摘要 首次以景天科多肉植物褐斑伽蓝叶片为外植体进行组织培养。建立植株再生系统后,研究了不同植物激素及其不同浓度配比对叶片分化的影响。试验结果表明:褐斑伽蓝叶片分化方式特殊,初代培养时外植体叶片分化困难,仅在附加6-BA(6-苄基腺嘌呤)1.5mg/L、IBA(吲哚丁酸)0.5mg/L的MS培养基上分化出绿色球状体。试管苗叶片与外植体叶片分化方式不同,在单独附加6-BA1.0~2.5mg/L或(萘乙酸)NAA0.1—1.0mg/L,以及6-BA与NAA不同浓度配比的培养基上出现了根、芽及愈伤组织等多种器官分化方式,且分化出的器官生长情况差异很大,可以根据不同的培养目的筛选不同的培养途径和适宜的培养基。 The study for the first time a high efficient regeneration system was established by using leaves of Kalanchoe tomentosa. Different concentration combinations of the growth regulators, 6-benzyl aminopurine (6-BA), naphthylacetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-D) were adopted to induce apparatus differentiation from leaves. The results showed that the leaves differentiation mode of Kalanchoe tomentosa is special. The green globoids were in- duced only on MS medium with additional 1.5 mg/L 6-BA and 0.5 mg/L IBA. When different hormones ( 1.0 2.5 mg/L 6-BA, 0. 1 - 1.0 mg/L NAA) were adopted to induce apparatus differentiation, the test-tube plants leaves could induce differentiation, but the differentiation type and the growth of differentiated organ are very different. The different culture approaches and the optimal medium can be screened according to different purposes.
