摘要
以牛血块为原料,采用超滤、加热、阴离子交换层析技术制备了牛血SOD。牛血块捣碎液1000ml可得比活为11324U/mg蛋白的SOD32mg,活性回收率为54.7%。以明胶为囊材,乳化法制备了SOD明胶微囊。其最佳制备条件为每1g明胶加入SOD20000U,乳化温度为60℃,乳化时间为5min,转速为250r/min。三批微囊的包封率分别为76.54%、81.32%、75.39%。以制得的SOD明胶微囊给小鼠灌胃,可显著性地抑制巴豆油引起的耳片肿胀。
Superoxide dismutase (SOD) was prepared from bovine blood block by ultrafiltration,heating and anion exchange chromatography. 32 mg of the purified SOD with specific activity of 11 324 U/mg protein (recovery 54. 7 % ) was obtained from 1000 ml of strapped fluid of blood block. The gelatin microcapsules of SOD were prepared with emulsion process using gelatin as load material.The optimal conditions for preparation were that SOD 20000 U in 1g of gelatin, emulsifying at 60℃for 5 min and stirring at 250 r/min. The rates of encapsulation of three batches of microcapsules were 76. 54 %, 81. 32 % and 75. 39 %, respectively. The SOD gelatin microcapsules given to ratS per os could significantly inhibit inflammatary edema of ears induced by croton oil.
