摘要
采用灰枣组培苗叶片、叶柄、茎段为试材,进行了体细胞胚胎发生的研究。结果表明,叶片、叶柄和茎段在MS+6-BA0.5mg/L+2,4-D0.5mg/L上的愈伤效果较好;MS+6-BA 0.5mg/L+2,4-D0.2mg/L+NAA 0.5mg/L比较适合叶柄胚性愈伤组织的形成;在MS+6-BA1.5mg/L+IBA0.2mg/L+GA2.0mg/L,附加蔗糖20g/L,AgNO32.0g/L的培养基上,灰枣体细胞胚胎的发生率最高;MS+6-BA0.5mg/L+NAA0.01mg/L培养基上的体胚萌发率较高,并且得到了再生植株。
Experiment was carried out with the leaf blade, petioles,stems of the jujube Huizao's vitro plantlets for studing the somatic embryogenesis. The results showed ..The callus tissue of leaves, stems and petioles growed better in MS media adding 6--BA 0.5mg/ L and 2,4--D 0.5 mg/L. The media of MS+6--BA 0.5mg/L+2.4--D0.2mg/L+NAA 0.5mg/L was better to the formation of petioles embryonic callus tissues. The regeneration rate of embryo with Huizao jujube was the highest in the media of MS+6--BA1.5mg/L+IBA0.2mg/L+GA2.0mg/L, plus sucrose 20g/l and AgNO3 2.0g/l. The germination rate of embryo was higher in MS+6--BA0.5mg/L+ NAA0.01mg/L, and the regeneration plantlets of Huizao jujube were obtained.
