摘要 目的探讨人类生物钟基因hClock mRNA及其蛋白在不同Dukes分期结直肠肿瘤中的表达,研究它们的表达与结直肠肿瘤的侵袭及转移的关系。方法采用原位杂交检测结直肠癌与相应癌旁组织中hClock基因mRNA的表达,并采用免疫组织化学检测相应标本中hClock基因蛋白产物(CLOCK蛋白)的表达。结果21例结直肠肿瘤中hClock mRNA弱阳性表达率47.62%,中或强阳性表达率52.38%,且与Dukes分期相关(P<0.05);CLOCK蛋白均呈中或强阳性表达。相应癌旁组织中hClock mRNA及蛋白呈弱阳性表达(P<0.01)。结论hClock基因可能与结直肠肿瘤的发生、发展及侵袭、转移有相关性。 Objective To explore the human circadian clock gene (hClock) mRNA and protein expression in colorectal tumors at different Dukes stages, and to study the relationship of their expression with eolorectal cancer invasion and metastasis. Methods The hClock gene mRNA expression in colorectal cancer and the corresponding adjacent tis- sues obtained from 21 patients were detected by In situ hybridization. The hClock gene protein product (CLOCK pro- tein) expression in corresponding specimens was examined by immunohistochemistry. Results The weak positive rate of hClock mRNA was 47.62% , and the medium or strong positive rate was 52.38% in 21 cases of colorectal cancer. The positive expression rate was related to the Dukes stages (P 〈 0.05). The CLOCK protein was medium or strong positive expression in all patients. The corresponding adjacent tissues hClock mRNA and protein expression were weak (P 〈0.01). Conclusion hClock gene may be related to the incidence of colorectal cancer, the development of invasion and metastasis.
