摘要
比较了不同体积摇瓶和16L(工作体积10L)搅拌式生物反应器中长春花(Catharanthusroseus(L.)G.Don)培养细胞生长和生物碱生成情况,发现在摇瓶中小规模放大培养,细胞生长和生物碱生成无显著差异,摇瓶体积不变而培养物体积增加时,细胞干重和生物碱生成均下降。反应器中培养细胞干重与摇瓶中相当,但生物碱含量大大下降。从而提示,培养环境的改变,特别是通气状况和剪切力是培养细胞从摇瓶到反应器放大过程中影响生物碱生成的主要因素。
The cell growth and alkaloid production of Catharanthus roseus (L.) G.Don cells cultured in the shake flasks with different volumes and in the stirred tank bioreactor (10 L) were compared. Cell dry weight and alkaloid production showed no significant difference in the small volume scaleup shake flasks. When more broths were added to a certain volume in the shake flask, both cell weight and alkaloid production were decreased. The maximum cell dry weight was similar between the cell cultures in the shake flask and the bioreactor, but the alkaloid production of cells was much less in the bioreactor. Gas regime and shear stress were recognized to be the main factors contributing the important effect on alkaloid production during the scaleup processes.
