摘要
目的研究三氧化二砷(As2O3)对人大肠癌裸鼠腹腔种植腹水生成的影响。方法将大肠癌Ls174细胞通过单独培养,与5-氟尿嘧啶(5-FU)、As2O3分别共培养后行腹腔内注射构建裸鼠腹水模型,通过单独和联合注入化疗药物,观察腹水生成情况以及检测腹水细胞中耐药基因谷胱甘肽硫转移酶酸性同工酶(GST-π)mRNA表达,比较三氧化二砷对大肠癌致腹水生成的影响。结果5FU和As2O3组裸鼠腹水生成明显减少,且生存期延长,耐药基因GST-π mRNA表达最低。结论三氧化二砷能够抑制或消除人大肠癌裸鼠腹腔种植及腹水的生成,不同程度地延长生存期并通过下调耐药基因GST-π mRNA表达提高对化疗药物的敏感性。
Objective To investigate the effect of arsenic trioxide on ascetic production in human colon cancer cell line Ls174. Methods Three kinds of ascites tumor-bearing mice model were constructed through intraperitoneal injection with Ls174 or well-prepared Ls174 incubated with 5-FU or As2O3. The effect of As2O3 on ascetic production was evaluated for every ascites tumor-bearing mice model, mice received 5FU, As2O3, or As2O3 combined with 5-FU intraperitoneally, respectively. The ascetic produetiogroln and survival time was observed. Meanwhile, the expression of glutathione-s-transferase π (GST-π ) in ascites was measured by RT-PCR. Results The prodwtion of aseites in group As2O3 with 5-FU is significatly inhibited, and the survival period is prolonged whaty more, the expression of GST-π mRNA was significant down. Conclusion As2O3 combined with 5- FU can inhibit and eradicate production of ascites and prolong the survival period of nude mice. As2O3 is able to partly reverse the resistance of the Ls174 cell of human cancer of colon, which may be related to the variation of GST-π.
