摘要
目的:建立黄芩药材的HPLC指纹图谱,并对其色谱条件进行优选。方法:采用梯度洗脱方法进行色谱分离,C18柱(4.6 mm×150 mm,5μm,Zirchrom Kromasil),流动相:乙腈与0.1%磷酸溶液进行梯度洗脱,检测波长278 nm,体积流量0.9 mL/min,柱温30℃。采用国家药典委员会开发的中药色谱指纹图谱相似度评价系统研究版(2004A)进行评价。结果:以黄芩苷峰为参照物峰,确定14个共有峰,确立了黄芩药材的HPLC指纹图谱。结论:该法精密度高、重现性好,所测组分均达到基线分离,可为黄芩药材的指纹图谱评价提供依据。
AIM: To establish the chromatogram fingerprints of Scutellaria baicalensis Georgi by HPLC and select the best chromatogram condition. METHODS: Agradient mobile system was applied in the experiment. The HPLC separation was performed on a Diamonsil C18 analytical column gradient eluted with acetonitrile-0.1% H3PO4 at the flow rate of 0.9 mL/min. The temperature of column was 30℃. The UV detection wavelength was at 278 nm. The chromatogram fingerprints were evaluated by the similarity evaluation software published by committee of codex. RESULTS: 14 co-possessing peaks were selected as the fingerprint peaks of Radix Scutellariae and the similarities between the chromatographic fingerprints and the herbal drugs were calculated by taking baicalin peak as the referential peak. The HPLC chromatogram condition of Scutellaria baicalensis Georgi was established. CONCLUSION: This method shows high precision, good repeatability and all of the contents are separated well, so it can be used as the means to assess the qualit of S. baicalensis Georgi.
