大鼠肝脏水通道蛋白8在胆道梗阻及再通时表达的变化 被引量:2

摘要 目的:研究胆道梗阻及再通后大鼠肝脏水通道蛋白8(AQP8)表达的改变及其意义。方法:40只大鼠按随机表法随机分为假手术组、胆道梗阻1周组、胆道梗阻2周组、胆肠内引流组。用免疫组织化学的方法对肝脏AQP8的表达进行检测。结果:免疫组织化学显示,AQP8阳性免疫反应产物位于肝细胞内和肝细胞膜上,假手术组AQP8表达尤其明显,胆道梗阻组大鼠肝脏表达AQP8均明显较假手术组减少(P<0.05),胆肠内引流组肝脏表达AQP8较胆道梗阻2周组增加(P<0.05)。结论:胆道梗阻导致肝脏AQP8表达减少,再通后表达增加,这种变化提示AQP8与肝脏胆汁分泌功能的改变有关。 To investigate the change of liver aquaporin 8 (AQP8) expression and its clinical significance during biliary obstruction and biliary decompression. Methods: Forty rats were divided into four groups randomly as control group, one week of bile duct ligation (BDL-1W) group, two weeks of bile duct ligation (BDL-2W) group, and biliary decompression (BDL-D) group. Immunohistochemistry was used to evaluate the expression of AQP8 in liver. Results: AQP8 immunoreactive products were observed on the membrane and inside of hepatocytes, especially in control group. IHC showed that the expression of AQP8 in the hepatocytes was decreased in both BDL groups when compared to the control group (P〈0.05), and AQP8 expression of BDL-2W group was lower than that in BDL-1W group. However, AQP8 expression was increased in BDL- D group compared to that of BDL-2W group (P〈0.05). Conclusion: The expression of AQP8 in the hepatocytes is decreasing as the result of bile duct ligation untill the ligation is removed, which indicates that there is a relationship between AQP8 expression and bile secretion.
