摘要
目的:探讨白藜芦醇(Res)对人激素非依赖性前列腺癌体外生长细胞PC-3的抑制作用及实现其抑制作用的可能途径。方法:按白藜芦醇浓度分为25,50,100和200 ttmoI/L 4个处理组和对照组(未加白藜芦醇),处理PC-3细胞不同时间后,倒置显微镜观察细胞形态;MTT法检测细胞的增殖抑制情况;免疫组化sP法分析Sur-vivin蛋白的表达。结果:与对照组比较,白藜芦醇处理组可使细胞明显变圆、体积缩小、脱壁细胞增多;MTT法检测显示,白藜芦醇对PC-3细胞的增殖有抑制作用,并呈时间-剂量依赖性,其中24 h组细胞存活率分别为86.2%,77.15%,60.89%和35.76%,48 h组细胞存活率分别为65.24%,59.76%,46.47%和27.77%,白藜芦醇处理组随着浓度的增加和作用时间延长,对PC-3细胞的增殖抑制作用增强,与对照组比较差异有统计学意义(P<0.05);免疫组化结果显示,Survivin蛋白在100 gmol/L白藜芦醇作用下较对照组表达明显减弱(P<0.01)。结论:白藜芦醇对体外生长的PC-3细胞有明显的抑制作用,下调Survivin蛋白表达而诱导前列腺癌细胞凋亡可能是其抑制肿瘤生长的机制之一。
To investigate the effect of Resveratrol on proliferation inhibition in human prostate carcinoma cells PC 3 in vitro and to explore the possible mechanisms involved. Methods: The PC-3 cells were divided into four treatment groups (25, 50, 100, and 200 μmol/L Resveratrol) and one control group. The morphological alterations of cells PC 3 after treated for different time were observed by using inverse microscopy; the inhibition of cell proliferation was detected by MTT assay; the protein level of Survivin in carcinoma cells was observed by SP immunohistochemistry. Results: After being treated with Resveratrol, PC 3 cells grew round and small obviously and the cell walls were isolated under the inverse microscope; Resveratrol could inhibit the proliferation of PC 3 cells, the inhibition of proliferation of PC-3 cells decreased in a time- and dose dependent manner, the survival rate of the four groups after 24 hours was 86.2%, 77.15%, 60.89%, and 35.76%,respectively, and the rate after 48 hours was 65.24%, 59.76%, 46.47%, and 27.77% ,respectively (P〈0.05). Compared with the control group, SP immunohistochemis-try showed the expression of Survivin protein in the PC 3 cells treated with 100 μ mol/L Resvera- trol was decreased (P〈0.01). Conclusion: Resveratrol could significantly inhibit the growth of cells PC-3 in vitro, the induction of apoptosis through down-regulating Survivin protein is probably one of it's molecular mechanisms on inhibition of tumor.
