右归丸对骨髓抑制小鼠骨髓细胞周期和凋亡的影响 被引量:13

摘要 目的观察右归丸对骨髓抑制小鼠外周血、骨髓细胞周期和凋亡的影响,探讨其可能造血调控作用机制。方法用全自动血细胞分析仪、白细胞计数法和流式细胞术(FCM)分别检测右归丸对骨髓抑制小鼠外周血、骨髓有核细胞数以及骨髓细胞周期和调亡的影响。结果右归丸中、高剂量均能明显升高外周血红细胞(RBC)、血红蛋白(Hb)、骨髓有核细胞(BMC)数,右归丸高剂量对血小板(PLT)和白细胞(WBC)有明显升高作用。右归丸低、高剂量组均能促进骨髓G0/G1期细胞向S期细胞以及S期细胞向G2/M期细胞的转化,从而导致G2/M期细胞比例明显升高,增殖指数(PI)也明显升高。中、低剂量组右归丸的骨髓细胞凋亡比例显著下降。结论右归丸可能通过促进骨髓细胞修复受损的DNA,加速通过G1/S和S期监测点,进行增殖和分化;抑制造血细胞的凋亡;调节造血细胞增殖与凋亡之间的平衡等机制,从而促进损伤骨髓造血功能恢复。 Objective To study the effect of You Gui Wan on peripheral blood cells, bone marrow cell cycle and apoptosis of myelosuppressed mice and to discuss its possible mechanism in hematopoietic regulation. Methods Automatic blood cell analysator, white blood cell counting and flow cytometry ( FCM ) were applied respectively in detecting the effect of You Gui Wan on peripheral blood cells, bone marrow nucleated cells, bone marrow cell cycle and apoptosis of myelosuppressed mice. Results Peripheral red blood cells, hemoglobin and bone marrow nucleated cells increased obviously by middle or high dose You Gui Wan ; high dose You Gui Wan had significant effect in raising the level of blood platelet and white blood cells ; low or high dose You Gui Wan promoted the transition of bone marrow cells from GO/G1 to S phase and that from S phase to G2/M phase, thus led to a higher cell proportion in G2/M phase as well as the proliferation index; middle or low dose You Gui Wan decreased bone marrow apoptosis gready. Conclusions You Gui Wan may promote the recovery of hemopoietic function of injured bone marrow cells by repairing the damaged DNA and it can accelerate GO/G - S phase and S - G2/M phase transition, activate the proliferation and differentiation, inhibit the apoptosis of hemopoietic cells and regulate the balance between their proliferation and apoptosis.
