Analysis of expression of dickkopf1 in two human glioma cell lines

目的检测Dickkopf1（DKK1）在人胶质瘤细胞株MGR3和UWR7中的表达情况。方法采用酶联免疫吸附实验，逆转录多聚酶链反应等方法对DKK1在人胶质瘤细胞株MGR3和UWR7中表达水平进行检测。结果酶联免疫吸附实验提示DKK1在MGR3和UWR7中均呈高浓度；逆转录多聚酶链反应提示DKK1基因在胶质瘤细胞株MGR3和UWR7中高表达，但在正常脑组织中表达不明显。结论DKK1在人胶质瘤细胞株MGR3和UWR7中高表达，提示DKK1可能在胶质瘤中的发生发展中起重要作用。

Objective To detect the expression of dickkopf1 （ DKK1 ） in human glioma cell lines MGR3 and UWR7. Methods The expression of DKK1 in human glioma cell lines MGR3 and UWR7 was detected by enzyme-linked immunosorbant assay （ELISA） and reverse transcriptase-polymerase chain reaction （RTff PCR）. Results ELISA demonstrated that the expression of DKK1 was both up-regulated in MGR3 and UWR7. RT-PCR showed that DKK1 transcripts was over-expressed in MGR3 and UWR7, which was absent in normol brain tissues. Conclusion DKK1 is over-expressed in glioma cell lines MGR3 and UWR7, which may play an important role in the initiation and development of glioma.
