摘要
[目的]研究半夏人工种子最佳种皮基质配比组合,为半夏人工种子技术规模化开发应用提供理论依据。[方法]以半夏组培小块茎为试验材料,选择7种人工种子包裹基质配比组合制成人工种子,在人工和自然环境下进行培养并测定人工种子萌发率,筛选出最佳的种子包裹基质配比组合。[结果]海藻酸钠(3.0%)+壳聚糖(2.0%)+活性炭(0.5%)+多菌灵(0.3%)+青霉素(0.02 mg/L)+苯甲酸钠(2.0%)+NAA(0.20 mg/L)+GA3(0.01 mg/L)+CaC l2(2.0%)为最佳种皮配比组合,萌发率达到84%。[结论]复合型半夏人工种皮具有抗菌、防腐、萌发率高等优点,可应用于生产实际中。
[ Objective] The research aimed to study the best seed coat matrix ratio combination of Pinellia ternate artificial seeds and provide the theoretical basis for development and large-scale application of Pinellia ternate artificial seeds. [ Methods] Using tubercle of tissue culture of Pinellia ternate as materials, selecting seven kinds of combination made into Pinellia ternate artificial seeds, determining artificial seed germination rate under the artificial and the natural conditions, the best combination of seeds coat matrix ratio was selected. [ Result] The best combination of seeds coat matrix was sodium alginate(3.0% ) + chitosan(2.0% ) + activated carbon (0.5%) + carbendazim(0.3% ) + penicillin(0.02 mg/L) + sodium benzoate(2.0% ) + NAA(0.20 mg/L) + GA3 (0.01 mg/L) + CaCl2 (2.0%), the germination rate was 84%. [ Conclusion] Multiple artificial seeds coat of Pinellia ternate have antibacterial, anti-corrosion, high germination rate and other advantages, which could be applied to practical production.
